研究简介:由于骨骼和免疫系统老化引起的改变,老年性骨质疏松性骨折的骨愈合延迟仍然是一个临床挑战。因则,临床上需要针对骨骼细胞和免疫系统缺陷的新型生物材料来有效治疗老年患者的骨损伤。锌 (Zn) 已显示出作为骨科植入物中可生物降解材料的诱人前景。为了解决老年骨损伤患者的骨愈合缺陷,我们联合四川大学华西口腔医院颌面外科罗恩主任/教授团队开发了一种可生物降解的锌基合金(Zn-2Cu-0.5Zr)。该合金机械力学性能较纯Zn和Zn-2Cu得到增强,屈服强度为198.7 MPa,极限抗拉强度为217.6 MPa。对骨髓间充质干细胞 (BMSC) 和 MC3T3-E1 细胞进行的细胞毒性试验表明,Zn-2Cu-0.5Zr 合金的浸提液样品没有表现出可观察到的细胞毒性作用。此外,Zn–2Cu-0.5Zr合金的浸提液通过调节炎症相关细胞因子的产生和调节巨噬细胞极化表现出显着的抗炎作用。进一步,改善的免疫骨微环境促进了BMSC的成骨分化。使用年龄相关性骨质疏松症大鼠模型测试证实了Zn-2Cu-0.5Zr合金满足服役的力学支撑性能要求,并加速了老年大鼠胫骨骨折的愈合过程。这些发现表明,新型Zn-2Cu-0.5Zr合金具有影响巨噬细胞免疫调节功能和促进老年骨质疏松症患者骨修复的潜力。

研究简介:开发合适的可降解金属材料对于骨质疏松性骨折治疗具有重要临床意义,但当前可降解金属在促骨再生修复方面仍存在不足,尤其是在骨形成和吸收之间缺乏良好的平衡。锌及其合金由于中等的腐蚀速率、良好的力学性能和锌离子的生物功能性,是应用于骨内固定植入物的潜在理想候选材料,但仍难以满足骨质疏松性骨折修复的需要,表面改性是根本途径。我们联合北京大学深圳医院陈英奇副研究员和南方医科大学第十附属医院张文泰副研究员在纯锌髓内钉表面设计了一种微/纳结构的金属-有机纳米棒(Zn-ZA&HEDP nano-stick)介导的磷酸锌杂化涂层,实现了成骨形成和破骨吸收之间的平衡。研究证实:改性的锌基髓内钉具有出色的成骨作用和抗破骨细胞分化的特性,在大鼠骨质疏松性骨折模型中表现出很好的促骨折愈合修复能力,这主要得益于该金属-有机/无机杂化涂层的生物功能活性组分及其特殊的微/纳结构、形貌与表面物理化学性质。研究为可降解金属的表面改性提供了新思路,并丰富了新型有机/无机杂化功能生物材料的理解和认识。

研究简介:锌及其合金由于其中等的腐蚀速率、理想的机械性能和锌离子潜在的生物功能性,在生物可降解骨植入物材料领域受到了广泛的关注。但是其腐蚀模式不均匀、成骨、抗炎和抗菌性能不足,难以满足骨植入体在临床应用中的综合性能要求。基于此,我们联合北京大学深圳医院陈英奇副教授等在纯金属表面设计了一种致密均匀的负载阿司匹林的有机金属水凝胶复合涂层,实现了锌基底的腐蚀调节、成骨、抗炎和抗菌功能。结果表明:改性的锌金属具有更均匀的腐蚀模式、更低的腐蚀速率;复合涂层促进成骨细胞增殖分化同时显示出抗炎性能;对大肠杆菌和金黄葡萄球菌均具有较高的抗菌率。该研究为可降解金属的表面改性提供了新思路,拓展了金属水凝胶材料在骨科领域的新应用,并加深了其机理性认识。

研究简介:神经血管介入血流导向装置(也称密网支架)使得脑部血管瘤瘤的治疗发生了革命性的变化。但目前的产品植入后患者仍必须服双抗治疗,由此带来了出血等不良事件风险的增高。表面改性对于血流导向装置的表面生物相容性及其临床服役功能的提升显得尤为重要和迫切。我们联合成都百瑞恒通医疗科技有限公司以及华中科技大学同济医学院附属协和医院胡学斌教授等在TiNi神经血管介入血流装置/密网支架表面通过化学浸涂的液相沉积方法获得了氧化钛共价固定柠檬酸纳米杂化功能改性层,以达到抗凝与促内皮的功能。研究证实柠檬酸分子有效地共价固接于预沉积的纳米氧化钛晶粒表面,该纳米杂化层具有独特、均一的纳米尺度形貌并体现出超亲水性表面物理化学性质。体外与体内评价结果表明改性后的样品显著阻碍血小板聚集和纤维蛋白原(第一凝血因子蛋白)的吸附,延迟凝血时间,同时促进再内皮化(自然抗凝表面)。以上功能主要得益于纳米杂化改性层组份柠檬酸及氧化钛的功能特性及其特殊的纳米结构特征与表面性质。该表面改性策略是最早使用功能分子杂化层以改善提高神经密网支架抗凝与促内皮化功能的报道,较当前其它方法研究具有超薄、稳定以及功能活性发挥高的优势,可望为神经血管介入血流导向装置表面改性提供新解决方案,并加深对新型纳米杂化材料在医学应用领域的理解和认识。
